•A new generation of protein-peptide co-assembling nanoformulation (PePCAN) platform was developed.•The PePCAN could efficiently deliver multiple therapeutic peptides to their cytosolic targets.•The PePCAN platform protected peptide medications from proteolytic degradation.•The PePCAN could release encapsulated peptides in a long-term manner.
