HPLC法测定葛根与粉葛中6个异黄酮类成分的含量

Abstract
目的:建立葛根及粉葛中3′-羟基葛根素、葛根素、3′-甲氧基葛根素、大豆苷、葛根素芹菜糖苷、葛根素-6″-O-木糖苷6个异黄酮类成分的含量测定方法.方法:采用HPLC法,Poroshell 120 EC-C18(150 mm×4.6 mm,4μm)色谱柱;以0.1%磷酸溶液-乙腈为流动相,梯度洗脱;流速为1.0 mL/min;检测波长为250 nm;柱温为25℃;进样量为10μL;采用聚类分析及主成分分析法进行数据分析.结果:3′-羟基葛根素、葛根素、3′-甲氧基葛根素、大豆苷、葛根素芹菜糖苷、葛根素-6″-O-木糖苷在相应的进样量范围内与峰面积的线性关系良好(r>0.9990),平均加样回收率为97.40%~101.93%,RSD为1.40%~2.85%.23批样品中6个成分含量依次为0.20~20.61 mg/g、4.28~76.10 mg/g、0.06~12.20 mg/g(部分样品未检出)、0.05~6.74 mg/g、0.72~13.18 mg/g、0.30~17.72 mg/g.聚类分析和主成分分析可用于区分葛根及粉葛样品.结论:该方法简便、准确、可靠,可为葛根、粉葛的质量标准建立及区分提供参考.
