Abstract
本研究旨在观察微小RNA(microRNA,miR)-199a-5p调控DDR1对人脑胶质瘤细胞增殖和迁移的影响. 一、材料与方法 1.材料:选择U251细胞作为研究对象,完成miR-199a-5p过表达U251细胞株.将其分作对照组(无转染的U251细胞),阴性对照组(转染空载体质粒U251细胞),试验组(转染miR-199a-5p成熟模拟物).通过实时荧光定量聚合酶链反应(polymerase chain reaction,PCR)法检测各组 miR-199a-5p 表达,以蛋白质印迹法(Western blot)法检测DDR1表达.采用细胞计数试剂盒(cell counting kit-8,CCK-8)法检测细胞增殖情况,以细胞划痕实验和Transwell迁移实验检测各组U251细胞迁移情况.并以Pearson相关性明确人脑胶质瘤细胞DDR1表达和细胞增殖、迁移的关系.
