•We assessed the pharmacological effects of semaglutide in atheroprone mice.•The model recapitulates central mechanisms related to VSMC phenotypic switching.•Semaglutide treatment reduced VSMC proliferation and vessel remodelling.•This effect was independent of GLP-1 receptor-mediated signalling in vascular cells.•Such mechanism of action may be important for the cardiovascular protection.
