•The sequence targeting tumor endothelial cell is introduced to the peptide gatekeeper.•The release of entrapped drugs is completely inhibited in the absence of stimuli.•The structure of peptide gatekeeper was converted by addition of bioreductant.•The entrapped drugs in nanocontainers are released on-demand.
