Abstract
优化铜藻中岩藻黄质提取工艺与分离纯化工艺.本研究以新鲜铜藻为原料,采用超声辅助有机溶剂萃取法提取,对岩藻黄质的提取工艺进行单因素实验与响应面法优化,随后采用6种不同大孔吸附树脂纯化岩藻黄质,进行静态吸附与解析动力学研究.结果表明:最佳提取工艺为:乙醇浓度95%,超声时间21 min,液料比38:1,提取次数为1次,所得岩藻黄质的提取得率为200.28μg/g(FW).拟一阶动力学模型能更好的诠释6种树脂吸附岩藻黄质的动力学行为,并结合动态吸附与解析结果筛选出最优性能的层析树脂为KP670型大孔吸附树脂(R2>0.99),一步柱层析使岩藻黄质质量分数从0.9%提高到31.07%.实验证明了该提取纯化铜藻中岩藻黄质的系列工艺是可行的.
