摘要

•JNK3 has become a promising drug target for Alzheimer’s disease.•Recent progress has been made in treating Alzheimer’s disease with JNK3 inhibitors.•This review is focused on small molecules targeting JNK3 and their mechanism of action.•The tight banding with hydrophobic picket Ⅰ is essential to improve JNK3 isoform-selectivity.•The formation of hydrogen bonds with Met149 is the key to improving inhibitory activity.
