•A long-acting formulation of oral buprenorphine (LYN-013) has been developed.•Buprenorphine was released over 7–10 days following a single oral dose.•LYN-013 produced sustained decreases in fentanyl self-administration.•LYN-013 was effective in male and female rhesus monkeys.
