The WNT signaling transduction pathway is a major regulatory force throughout the development. Cells will respond to Wnt ligands in a context dependent manner to mediate cell proliferation and differentiation. WNT/β-catenin signaling pathway has been revealed to participate in both normal and cancer stem cell maintenance. With recognition of the importance of cancer stem cell, the regulation of WNT/β-catenin signaling pathway continuously attracts the attention of research and medical therapies. Here, we describe the canonical and non-canonical WNT/β-catenin signaling pathway, the correlation and importance of certain phosphorylation events on β-catenin. Finally, we discuss WNT/β-catenin in stem cells.
