•Histone chaperones have been only found in eukaryotes.•We identified the EB virus tegument protein BKRF4 as a bona fide histone chaperone.•We revealed the molecular basis of its recognition of histones H3–H4 and H2A–H2B.•The conserved BKRF4 motifs D/EL and DEF/Y/W target H3–H4 and H2A–H2B, respectively.
