•The ESI-MS/MS spectra of N-benzylated cathinones are uninformative.•Structural elucidation was achieved through derivatization and LC–MS/MS analysis.•The gas-phase fragmentation behavior of the derivatized cathinones was studied.•The method facilitates the differentiation of isomeric synthetic cathinones.•The method was applied to urine.
