miR-211-5p在局灶性脑缺血再灌注损伤大鼠模型中表达分析

Abstract
目的 探讨大鼠局灶性脑缺血再灌注(MCAO/R)损伤模型大鼠脑组织miR-211-5p表达情况.方法 取SPF级成年大鼠40只,分为假手术组(Sham组)、MCAO/R+溶剂组(Vehicle组)、MCAO/R+miR-211-5p组(Mimic组)、MCAO/R+anti-miR-211-5p组(Antagomir组),每组10只.评价各组大鼠神经功能缺损评分,检测各组大鼠脑梗死体积及miR-211-5p、脑源性神经营养因子(BDNF)蛋白表达水平.结果 Vehicle组存活率低于Sham组,Longa评分升高,miR-211-5p处理后存活率升高,Longa评分降低(P<0.05).Mimic组脑梗死体积百分比较Vehicle组降低,An-tagomir组脑梗死体积百分比高于Mimic组(P<0.05).与Sham组和Vehicle组比较,Mimic组miR-211-5p表达量升高,Antagomir组表达量降低,Antagomir组miR-211-5p表达量低于Mimic组(P<0.05).Mimic组BDNF表达量显著高于其他3组,Antagomir组BDNF表达量最低(P<0.05).结论 miR-211-5p表达与大鼠MCAO/R损伤相关.
