Determination of gamma radiation sterilization dose of nano-chitosan/cellulose microbial composite for splinting periodontal

Tooth mobility is indicated by over mobility of teeth. If not treated properly, tooth mobility finally causes lost of teeth. To prevent the worstness of tooth mobility, the splinting materials which can immobilize the movement of the tooth are needed. Nano-chitosan/cellulose microbial (CHNP/CM) composite is potentially used impant material for the splinting material to manage tooth mobility. The requirement of impant material is it should be sterile. Gamma radiation is used for sterilization of CHNP/CM composite. The International Standard ISO 11137-1 method 1 was used to determine the radiation sterilization dose of the CHNP/CM composites. To calculate the absorbed dose of the sample, Harwell Dosimeter was used. Sterility Assurance Level of 10−6 and sample item portion (SIP) of 1 were selected. One hundred (100) samples were irradiated with gamma-ray using determined verification dose. The results showed that the average bioburden of the composites was 16.7 CFU, and it was used to obtain verification dose. The verification dose for 16.7 CFU is 5.8 kGy. The absorbed doses were found ranging from 5.57 and 5.81. These doses comply with the arithmetic mean of the highest and lowest doses (90% to 110 % of verification dose). From the test results of sterility of 100 CHNP/CM composites samples showed that only one out of 100 samples was non-sterile. Since less than 2 samples were non-sterile, the verification dose was accepted. From Table 1, the sterilization dose of 100 CHNP-CM composites Sterility Assurance Level (SAL) of 10−6 and average bioburden 16.7 CFU is 18.4 kGy. It can be concluded that CHNP/CM composites with SAL 10−6 can be radiation sterilized with a dose of 18.4 kG.
