牦牛TLR-4基因3'-UTR区扩增克隆及相关miRNAs生物信息学分析

Abstract
本试验采用构建DNA混池和测序的方法,对西藏、青海341头牦牛TLR-4基因3'-UTR区单核苷酸多态性(SNP)位点进行筛选,然后用DNAMAN 8.0软件进行序列对比分析,并通过TargetScan 7数据库预测牦牛TLR-4基因3'-UTR区的微小RNAs (miRNAs)的可能靶定.结果 显示,牦牛TLR-4基因3'-UTR区可能存在2个SNP位点,分别为A38UG和A382G,通过TargetScan7数据库在线预测到牦牛TLR-4基因3'-UTR区共18个miRNAs可能靶定.本试验为牦牛TLR-4基因3'-UTR区可能存在的A380G和A382G突变位点与所预测的miRNAs之间靶向验证提供了初步理论根据,提示可通过分子水平进行抗病育种提高牦牛群体免疫力.
