miR-495-3p靶向IL-6R抑制宫颈癌细胞的增殖和转移的机制研究

Abstract
目的:探讨miR-495-3p和白细胞介素6受体基因(IL-6R)在宫颈癌细胞增殖和转移中的作用.方法:实时荧光定量PCR(RT-qPCR)和Western blot检测人宫颈永生化细胞H8和宫颈癌细胞(SiHa、HeLa、caski)中miR-495-3p和IL-6R表达水平.将SiHa细胞分为miR-NC、miR-495-3p、si-NC、si-IL-6R、miR-495-3p+pcDNA-NC、miR-495-3p+pcDNA-IL-6R组.细胞计数试剂盒(CCK-8)、Transwell实验分别检测SiHa细胞的增殖和转移能力.Western blot检测细胞周期素D1(CyclinD1)、基质金属蛋白酶2(MMP2)、MMP9蛋白表达.双荧光素酶报告实验、Western blot确定miR-495-3p对IL-6R的靶向调控作用.结果:与H8细胞比较,宫颈癌细胞中miR-495-3p表达降低,IL-6R表达升高(P<0.05).过表达miR-495-3p或抑制IL-6R表达后SiHa细胞CyclinD1、MMP2和MMP9表达降低,细胞增殖和转移能力降低(P<0.05);过表达IL-6R可逆转miR-495-3p对SiHa细胞增殖、转移以及其相关蛋白表达的影响(P<0.05).IL-6R是miR-495-3p的靶基因,miR-495-3p负调控IL-6R表达.结论:miR-495-3p通过靶向IL-6R抑制宫颈癌细胞的增殖和转移,提示miR-495-3p在宫颈癌中具有抑癌作用.
