养肝益水颗粒对自发性高血压大鼠血浆AngⅡ、AT1R含量及肾脏AngⅡ、AT1R、TRPC6蛋白表达的影响

Abstract
目的 观察养肝益水颗粒对自发性高血压大鼠(SHR)血浆AngⅡ、AT1R含量及肾组织Ang Ⅱ、AT1R、TRPC6蛋白表达的影响,探讨其可能的肾脏保护机制.方法 选取SHR40只,高脂饲料喂养4周后,随机分为SHR+Y组(养肝益水颗粒组)、SHR+B组(贝那普利组)、SHR+Y+B组(养肝益水颗粒+贝那普利组)、SHR组,另选周龄、性别配对的WKY大鼠10只;给药按照SHR+Y组给予养肝益水颗粒2.7g生药/(kg·d),SHR+B组按照贝那普利0.9mg/(kg·d)灌胃,早8~10时灌胃,SHR+Y+B于早8~10时、晚18~19时两次分别予以养肝益水颗粒、贝那普利灌胃.SHR组及WKY组均给予2ml生理盐水于早8-10点灌胃;治疗4周后测定血清AngⅡ、AT1R含量;Western blot法测定各组肾脏组织AngⅡ、AT1R、TRPC6蛋白表达水平.结果 与SHR组相比,SHR+Y组、SHR+B组及SHR+Y+B组血清Ang Ⅱ、AT1R含量,肾组织AngⅡ、AT1R、TRPC6表达水平均显著降低(P<0.01);SHR+Y+B组降低血清AngⅡ含量显著优于SHR+Y、SHR+B组(P<0.01),SHR+Y+B组降低AT1R、TRPC6表达水平显著优于SHR+Y组(P<0.01).结论 养肝益水颗粒可通过降低SHR血清Ang Ⅱ、AT1R含量及肾脏组织内Ang Ⅱ、AT1R、TRPC6蛋白表达水平,发挥肾脏保护作用,联合贝那普利则疗效更优.
