Effect of Buzhong Yiqi Decoction(补中益气汤) on Memory Ability of SCH Rats Based on microRNA132/212 Regulating BDNF/Trkb-ERK1/2 Signaling Pathway

Abstract
目的 观察补中益气汤及补中益气+二陈汤对SCH大鼠海马microRNA132/212-BDNF/Trkb-ERK1/2信号通路的影响,探讨其对SCH脑损伤作用机制的研究及有效方剂的筛选.方法 SPF级Wistar雄性大鼠40只,随机取8只做假甲状腺全切术作为空白组,其余采用甲状腺全切+ LT4替代的方法造模,分为模型组、补中益气汤组、补中益气+二陈汤组、左甲状腺素组,每组8只,空白组和模型组给等量生理盐水,其他各组给予对应的干预因素,1次/d,共28 d.光镜下×400倍放大观察HE染色海马神经元形态,电镜下观察海马神经超微结构;放免法测定血清TT4免疫化学发光法检测TSH;QT-PCR法测定各组大鼠海马miRNA132、miRNA212;RT-PCR法测定各组大鼠海马BDNF及磷酸化TrKB、ERK1/2表达.结果 与空白组比较,模型组TSH水平显著增高,BDNF、p-TrKb、p-ERK1、p-ERK2、miRNA132、miR-NA212 mRNA积分光密度值减低;与模型组比较,各治疗组TSH水平显著降低,海马神经元结构明显恢复,BDNF、p-Trkb、p-ERK1/2、miRNA132、miRNA212 mRNA积分光密度值显著增加(P<0.05,P<0.01);与补中益气汤组比较,补中益气+二陈汤组在改善BDNF、p-Trkb、p-ERK1/2、miRNA132、miRNA212 mRNA积分光密度值更优(P<0.05).结论 补中益气+二陈汤可改善SCH大鼠记忆能力,其机制可能与上调大鼠海马中miRNA132、miRNA212水平,进而调节BD-NF及其受体Trkb表达,激活细胞外蛋白调节激酶ERK1/2有关.
