Abstract
目的 建立乙型脑炎病毒微滴式数字PCR (droplet digital PCR,ddPCR)检测方法.方法 根据乙型脑炎病毒基因的NS3序列设计特异性引物和TaqMan探针,用于ddPCR和实时荧光定量PCR(real-time quantitative PCR,qPCR).优化ddPCR的退火温度、引物和探针浓度后,比较ddPCR和qPCR灵敏度和重复性,并用多种虫媒病毒进行特异性验证.结果 该方法特异性良好,灵敏度可以达到16.1 copies/μL,重复性良好.结论 本研究建立的ddRT-PCR能够高度敏感地检测乙型脑炎病毒核酸,将该方法用于临床乙型脑炎病毒感染的检测,可提高检测的准确性.
