Abstract
目的 应用超高效液相色谱-四级杆-飞行时间高分辨质谱(UPLC-Q-TOF-MS/MS)技术分析防风与其伪品水防风化学成分的差异,结合薄层色谱法鉴别防风药材真伪.方法 以0.1%甲酸乙腈(A)-0.1%甲酸水溶液(B)为流动相,梯度洗脱;流速:0.24 mL·min-1;进样量:1μL.质谱条件为电喷雾离子源(ESI),在负离子模式下对色谱流出物进行检测,检测范围(m/z) 100~1500.以6''-O-apiosyl-5-O-Methylvisammioside为对照品,对防风药材和水防风药材进行薄层色谱鉴别.结果 在鉴定得到的29个化合物中,防风有22个化学成分,水防风有23个化学成分.负离子模式下筛选得到6"-O-apiosyl-5-O-Methylvisammioside、花椒毒素、divaricatol、3'-O-2-乙酰亥茅酚、白花前胡乙素、3-(((3R,4R,5S)-6-((((3R,4R)-3,4-dihydroxy-4-(hydroxymethyl)tetrahydrofuran-2-yl)oxy)methyl)-3,4,5-trihydroxytetrahydro-2H-pyran-2-yl)oxy)-5-hydroxy-2,2,8-trimethyl-3,4-dihydro-2H,6H-pyrano[3,2-g]chromen-6-one及其异构体7个差异化合物.薄层色谱中水防风药材和对照品在相应位置显示相同颜色的荧光斑点,可鉴定为防风伪品.结论 以UPLC-Q-TOF-MS/MS技术结合薄层色谱法建立了防风和水防风化学成分的鉴别方法,该方法简便、快速、准确,可为防风药材的质量评价及药效物质基础研究提供参考.
