THE ROLE OF LNCRNA NORAD AND MIR-26A-5P IN THE FORMATION OF KELOID

Objective: The present study aims to explore the mechanism of action of LncRNA NORAD and miR-26a-5p in the proliferation, migration and invasion of human keloid fibroblasts. Methods: the ciRT-PCR method was used to detect the expression of NORAD and niiR-26a-5p in normal skin tissues and keloid tissues; si-NC, si-NORAD, miR-NC, miR-26a-5p mimics, si-NORAD and anti-miR-NC, si-NORAD and anti-miR-26a-5p were transfected into keloid fibroblasts (si-NC group, si-NORAD group, miR-NC group, miR-26a-5p group, si-NORAD+anti-miR-NC group, si-NORAD+anti-miR-26a-5p group); the CCK-8 method was used to detect cell viability; the Transwell chamber experiment was used to detect cell migration and invasion; the dual-luciferase reporter gene experiment was used to detect the target relationship between NORAD and miR-26a-5p; western blot was used to detect the protein expression of Ki-67, MMP-2, and MMP-9. Results: Compared with normal skin tissue, the expression level of NORAD in keloid tissue increased (P<0.05), while the expression level of miR-26a-5p decreased (P<0.05); compared with the si-NC group, the cell viability of the si-NORAD group reduced ( P<0.05 1, the number of migrating and invading cells significantly reduced (P<0.05), and the protein levels of Ki-67, MMP-2 and MMP-9 were (P<0.05); NORAD could target miR-26a-5p; compared with the miR-NC group, the cell viability of the miR-26a-5p group reduced (P<0.05). The number of migrating and invading cells significantly reduced (P<0.05), and the protein levels of Ki-67, MMP-2 and MMP-9 reduced (P<0.05); compared with the si-NORAD+anti-miR-NC group, the cell viability of the si-NORAD+antimiR-26a-5p group increased (P<0.05), and the number of migrating and invading cells significantly increased (P<0.05). The protein levels of Ki-67, MMP-2, MMP-9 increased ( P<0.05 ). Conclusion: Interfrrence with the expression of NORAD could reduce the proliferation, migration and invasion of keloid fibroblasts by up-regulating the expression of miR-26a-5p.
