EXPRESSION DIFFERENCES OF TREG AND TH2 CELL SUBSETS, RELATED CYTOKINES AND TRANSCRIPTION FACTORS IN PERIPHERAL BLOOD OF PATIENTS WITH ALLERGIC RHINITIS

Objective: To explore the differences of Treg and Th2 cell subsets, related cytokines and transcription factors in the peripheral blood of patients with allergic rhinitis. Methods: A total of 46 patients with allergic rhinitis who were treated in the Department of Otorhinolaryngology of our hospital from April 2018 to March 2020 were randomly selected as the study group. During the same period, 46 healthy subjects in our hospital were selected as the control group. The gender, age, height and other basic data of the study group and the control group were compared. The percentage of Th2 cells, CD4+, CD25+ Treg cells and FOXPCD4+ CD25+ Treg cells were measured by attune NXT . flow cytometry. The levels of transforming growth ,factor-beta 1 (TGF-beta 1), interleukin-4 ( IL-4) and interleukin-5 (IL-5) were detected by enzyme-linked immunosorbent assay (ELISA). Real-time fluorescent quantitative PCR was used to determine the expression levels of the peripheral blood transcription factor forkhead transcription factor P3 (FOXP3) and type IV zinc linger protein 3 (GATA3) that can bind to the DNA sequence AIT (GATA) AIG in each group. Results: Compared with the control group, in the study group the percentage of Th2 cells and the levels of IL-4, IL-5 and GATA3 mRNA were significantly increased, while the percentage of Ca-4(+) CD25+ Trek cells, FOXP3+ CD4+ CD25+ Treg cells, TGF-beta 1 levels and FOXP3 mRNA were significantly decreased (P<0.01). Conclusion: Patients with allergic rhinitis have obvious Treg/Th2 imbalance, which may play an important role in the occurrence and development of allergic rhinitis.
