阿哌沙班中基因毒性杂质的LC-MS/MS检查

Abstract
目的:建立LC-MS/MS法同时测定阿哌沙班中苯肼类、苯胺类、硝基苯类与氯代烷烃类5个基因毒性杂质.方法:采用Agilent C18色谱柱(150 mm×4.6 mm,5μm),以0.1%甲酸铵-乙腈(90∶ 10)为流动相A、乙腈为流动相B,线性梯度洗脱,质谱电喷雾正离子化多反应监测各基因毒性杂质,氯雷他定内标法定量测定.结果:5个基因毒性杂质均在20(50) ~500 ng·mL-1范围内线性关系良好;检测限和定量限分别低达10和20 ng· mL-1;平均回收率均为90.7%~107.2%.结论:建立的LC-MS/MS测定法专属性好、灵敏度高,适用于阿哌沙班中苯肼类、苯胺类、硝基苯类与氯代烷烃类5个基因毒性杂质的检查与控制.
