Strategy for optimized expression of beta-barrel proteins from pathogenic Gram-negative bacteria to the outer membrane of Escherichia coli

Outer membranes (OM) of Gram-negative bacteria often contain essential components for pathogen-host interactions, infection and disease progression of pathogenic species. Such virulence factors include, but are not limited to, β-barrel outer membrane proteins and modified lipids, such as lipopolysaccharide (LPS). Investigating β-barrel outer membrane proteins in a more native environment can often be challenging in vitro due to difficulties in expression to and purification from the membrane. Studying virulence factors of pathogenic bacteria in vivo is also complicated by the need for higher biosafety lab facilities.
