A Carry-On Kit Containing Electrospun Nanofibrous Affinity Membranes by Surface Grafting Phenylboronic Acid for Quantitative Enrichment of Nucleotides in Urine

It has great potential to develop a carry-on kit for quantitatively enrich nucleotide biomolecules in biological urine. In this study, the electrospun nanofibrous membranes were fabricated on and tailored along the circular coverslips for 6-well orifice plates with specific surface area. After hydrophilic modification by plasma, the orifice membranes were grafted with phenylboronic acids as functional groups by EDC-NHS reaction. Various morphological characterization showed effective introduction of quantitative phenylboronic acid groups onto the surface of the membranes. The sampling and chromatographic conditions were optimized for quantitative enrichment of nucleotides in urine by the affinity membranes in the orifice plate well. The carry-on kit containing the affinity membranes exhibit considerable sensitivity (LOD 5.0 ng mL(-1)) and quantification (recoveries: 78.5 %-93.1 %) for spiked nucleotides in urine. It is low-cost, convenient, and has great potential for developing standard operation procedure of quantitative enrichment of nucleotides in complex uric matrix with controllable adsorption capacities.
