Optimization of GBE extracting technology and effect of GBE on expression of Bcl-2 mRNA in Hela cell

Objective To study the extraction technology optimization of flavonoids of Ginkgo bilobal and the anti tumour of the extraction of Ginkgo bilobal(GBE).Methods Different methods of alkali,alcohol and acetone were used.The ratio of solid to liquid and extrating time were optimized.Hela cell was cultured in RPIM 1640 without calf and in GBE as revulsant.After 24h,the expression of Bcl 2 mRNA was measured.Results The result showed that the best process was that alcohol concentration was 70%,the ratio of solid to liquid 1∶3,and reflux for 2h.After 24h,the expression of Bcl 2 mRNA by RT PCR were found decreased by GBE.Conclusion The best process is that the concentration of alcohol is 70%,the ratio of solid to liquid 1∶3,and reflux for 2h.GBE can decrease the expression of Bcl 2 mRNA.And it may has anti tumour effect.
