Effect of high glucose on mouse podocyte adhesion and its possible mechanism

Objective To investigate the effect of high glucose(HG)on mouse podocyte adhesion and its possible mechanism.Methods Conditionally immortalized mouse podocytes were treated with normal glucose(NG),HG and mannitol control.Cell attachment was measured by fluorescence and centrifugation cell adhesion assays,respectively;Expression of α3β1 integrin mRNA and protein was examined by real-time PCR and western blot,respectively.Results Compared to NG, HG induced a significant reduction in adherent podocytes to basement membrane protein complex in a time-dependent manner(P0.05);HG also caused significantly time-dependent decreases inα3 andβ1 integrin expression,both at protein and mRNA level,compared with NG(P0.05).Conclusion HG can remarkably inhibit mouse podocyte adhesion,which might be related to downregulation ofα3β1 integrin.
