Inhibition of rosiglitazone on NE and PMA induced growth-promoting effects in rat cardiac myocytes in vitro

AIM: To investigate the inhibitory effects of rosiglitazone(RSG) on noradrenalin(NE)-induced cardiac myocyte hypertrophy and the mechanism involved. METHODS: NE has been shown to be a potent stimulator for neonatal rat ventricular myocyte hypertrophy in vitro. We used the activator of PPAR γ(RSG), the inhibitor of protein kinase C(PKC) chelerythrine (che) to influence protein content.3H-leucine incorporation and activity of PKC were measured on cardiac myocytes stimulated by Chronic NE or PMA(the activator of PKC). RESULTS: After 2-d culture, RSG not only inhibited the inducing effects of NE and PMA on the elevation of protein content and 3H-leucine incorporation on cardiac myocytes, but also decreased the enhancing effects of NE and PMA on the activity of PKC in myocytes. RSG and che decreased the enhancing effects of NE on c-fos protein expression. CONCLUSION: Inhibitory effects of RSG on NE inducing growth-promoting in myocytes are associated with PKC and c-fos pathway.
