The effect of intense pulsed light(IPL) on MMP-1 and TIMP-1 expression in human culture fibroblasts

Objective To observe the effect of intense pulsed light(IPL) on expression of MMP-1 and tissue inhibitor metalloproteases-1(TIMP-1)in culture human fibroblasts and investigate biological mechanisms of photorejuvenation.Methods Cultured human fibroblasts were treated by IPL radiation(20,25,30 J/cm2),MMP-1 and TIMP-1 in the supernatant liquid were measured by ELISA after 24h and 48h.Results After 24h,the concentration of MMP-1 and TIMP-1 in the supernatant liquid increased a little compared with controls at 25、30 J/cm2;after 48h,the concentration of MMP-1 in the supernatant liquid increased in all three energy groups,but no significant difference among every group,however,TIMP-1 increased with increase of energy.Conclusion After treatment by IPL,the expression of MMP-1 and TIMP-1 of fibroblasts increased,but TIMP-1 increased far more than MMP-1.This indicate that biological mechanism of photorejuvenation of IPL associate with the increase of TIMP-1 which fibroblasts produce.
