Abstract
Objective To observed the therapic apoptosis effection of Hepatocellular Carcinoma-Targeting Adenovirus Containing r-caspase3 gene which on AFP expressed hepatocellular carcinoma. Methods The AFP expressed cells HepG2,7721,normal hepatocyte L-02 and breast cancer cells MDA-MB-231 were infected by recombinant Adenovirus Ad-r-caspase3,Ad-EAFPPALB/r-caspase3 and Ad-PALB/r-caspase3, the O.D figure were detected by SRB dyeing,and the apoptotic figure were tested by FCM. Results The sensitivity after infection with Adenovirus on HepG2 and 7721 showed: Ad-EAFPPALB/r-caspase3 Ad-PALB/r-caspase3 Ad-r-caspase3;the data of FCM showed:the recombinant Adenovirus of Ad-EAFPPALB/r-caspase3 has very strong apoptotic specificity on AFP expressed cell. Conclusion The Recombinant of Hepatocellular carcinoma-targeting Adenovirus Containing r-caspase3 gene has apoptotic specificity on AFP expressed cell and the more stronger expression the more apoptotic specificity.
