The detection of five SCFA in human's feces by HPIEC

A method to determine the short-chain fatty acids in human feces was established,which is highly efficient,rapid,as well as accurate.The five kinds of short-chain fatty acids which existed in human feces(SCFA: acetic acid,propionic acid,isobutyric acid,butyric acid,isovaleric acid) were separated and determined by ion-exclusion chromatography.Under a temperature of 50 ℃,using phosphoric acid(volume fraction is 0.1%) as the mobile phase,the extracted samples have been separated by a Shodex Rspak KC-811 ion exclusion column successfully.Meanwhile,those organic acids in them have been detected by a UV detector under a wavelength of 210 nm.Concentrations of the measured components and their peak areas both showed good linear relationships between 2.5 and 500 mg / L with correlation coefficient R2of ≥ 0.999,and the RSDs of mass concentrations of all the components were within the range of 2.3% ~ 3.9%.Average recoveries were from 85.5% to 105.7%.The method is simple,effective and accurate in the determination of several major SCFAs in human feces.Thus,it laid a strong foundation for the exploration of the physiological mechanisms which SCFA functioned in the normal guts.
