Effects of 5-Aza-CdR and low concentration of cisplatin on non-small cell lung cancer cells

Abstract
Objective To investigate the combination therapy of 5-aza-2'-deoxycytidine( 5-Aza-CdR) and low concentrations of cisplatin( DDP) on non-small cell lung cancer A549 cells. Methods A549 cell was treated by 1μmol/L DDP,10μmol/L 5-AzaCdR and combination of two drugs. MTT test was used to determine the change of cell proliferation. The cell cycle and apoptosis index were analyzed by flow cytometry. The morphology of cell apoptosis was analyzed by Hoechst 33258 staining. Results 1μmol/L DDP combined with 10μmol/L 5-Aza-CdR inhibited A549 cells proliferation more prominently than 1μmol/L DDP. The apoptisis rate of A549 treated by 5-Aza-CdR and 1μmol/L DDP was higher than that treated by 1μmol/L DDP. 1μmol/L DDP combined with 10μmol/L 5-Aza-CdR blocked cell cycles at G1/G0phase and cells proliferation index reduced. The morphology of A549 cells apoptosis were changed more significantly in combined group than in 1μmol/L DDP group. Conclusion The apoptosis rate of non-small cell lung cancer A549 cells could be increased by combination of 5-Aza-CdR and low concentrations of DDP.
