Exploration of alfa-amylase Inhibitor producing marine actinobacteria and evaluation of its antioxidant and antimicrobial activities –In vitro study

The purpose of this study is to explore the α-amylase enzyme inhibitor producing of actinobacteria from salt pan soil sample collected from coastal areas of Kothapattanam, Ongole, Andhra Pradesh, India. Among 28 colonies, only 3 acclimatized actinobacterial isolates (VITISB-3, VITISB-9 and VITISB-23) showed potency during primary screening technique. The isolates were inoculated in production media for secondary screening and bioactive compound extraction using different solvents of varying polarity. The ethyl acetate extracts of VITISB-23 and VITISB-3 isolates showed significant α-amylase inhibition activity while VITISB-9 showed less activity. These isolates were further screened for antimicrobial activity and free radical scavenging antioxidant potential. Among these isolates, VITISB-23 and VITISB-3 showed good antimicrobial activity while the other isolate, VITISB-9 showed moderate inhibition activity. VITISB-23 showed antimicrobial activity against all the pathogenic test organisms used in the study. In DPPH antioxidant assay, the isolate VITISB-23 showed a 96% free radical scavenging property, where VITISB-3 showed more than 80%. Based on these results we concluded that VITISB-23 possess potential biological activity and can be further studied for the isolation of new anti-diabetic compound and also for future commercial prospects in pharmaceutical industry for the formulation of medicinal drugs.
