Abstract
The antiviral activity of cycloferon with respect to influenza A(H3N2)/Aichi/1/68 virus was studied in vitro on MDCK and FLAC cell cultures in the «treatment-and-prophylactic» drug administration scheme. Two drug concentrations (50 and 100 mg/mL) were considered in time points of cycloferon exposure with cell cultures eitgher simultaneously with the introduction of influenza A virus (H3N2) or 4 and 24 h after infection (the multiplicity of infection was 0.01 MOI/TCID 50 per cell). The following indicators of drug action were considered: hemagglutinin titers, neuraminidase RNA expression level, virus RNA expression level, level of accumulation of influenza A(H3N2) virus (infectious activity), and the drug protection index. The introduction of cycloferon at concentrations of 50 and 100 mg/mL into the FLAC culture infected with the influenza A(H3N2) virus at a minimal multiplicity of infection (0.01 MOI/TCDI 50 /cell) in 24 h decreases the level of hemagglutinin by half ( p < 0.05), reduces neuraminidase level 1.6 – 5.2 times ( p < 0.05), and reduces the number of viral RNA copies 12 – 15.3 times ( p < 0.05). The obtained results confirm the in vitro antiviral effect of cycloferon as manifested by inhibition of the adhesive and reproductive activity of influenza A(H3N2) virus, which ensures a high (51.3%) drug protection index.
