Abstract
On the 7th day after the modelling of periodontitis, there was a decrease in the relative number of macrophages in peritoneal washout, but with an increase in the number of adhered cells. A decrease in the macrophage adhesive capability was established on the 14th day of experiment only. The phagocytic activity of neutrophils was reduced in the animals with periodontitis as manifested by decrease in the phagocytic and opsono-phagocytic indices by 40.1 and 42.9%, respectively, on the 7th day of experiment ( p < 0.05 – 0.001) and phagocytic index by 31.5% on the 14th day. The amount of phagocyting neutrophils did not differ significantly from the same value in naive animals on the 21st day. The inhibition of phagocyte absorption capability was not established in the animals with periodontitis. The administration of indolicidin analogs to the animals with periodontitis at the dose 500 μg/kg intraperitoneally once within 7 days had a stimulatory influence on both the relative amount of macrophages in the peritoneal washout (for indolicidin No. 7, by 24.0% and for indolicidin No.8, by 20.0%, p < 0,001) and the percentage content of adhered macrophages: beginning witgh the 14th day of experiment, the increase of percentage content of adhered cells was 21.7 and 17.1% upon the administration of indolicidin No. 7 and indolicidin No. 8, respectively, as compared to the control group ( p < 0.05). The investigated indolicidin analogs produced a stimulatory effect on the phagocytic activity of neutrophils during all the time of observation, as manifested by significant increase in all the investigated indices: phagocytic index by 43.0 – 50.0% ( p < 0.001), phagocytic number by 12.6 – 50.0% ( p < 0.05 – 0.01) and opsono-phagocytic index by 77.5 – 173.5% ( p < 0.05 – 0.01). The stimulatory effect of indolicidin No. 7 on the phagocytic activity of neutrophils of peripheral blood and the adhesive capability of peritoneal macrophages was higher than that of indolicidin No. 8. In particular, only indolicidin No. 7 caused an increase in phagocytic number and relative amount of adhered peritoneal macrophages on the 21st day, and the growth of opsono-phagocytic index was also more significant after indolicidin No. 7 administration.
