Abstract
The embryotoxic effects of a new normotymic drug based on the complex of lithium citrate, polymethylsiloxane, and aluminum oxide (hereinafter referred to as the lithium complex) have been studied in the antenatal and postnatal periods of development. It was established that intragastric administration of the lithium complex in doses of 400 and 2000 mg/kg once a day from the 1st to 19th and from the 6th to 20th day of pregnancy led to a number of dose-dependent embryotoxic effects in rats, which were manifested in both antenatal and postnatal periods. In the antenatal period of development of the offspring of animals, the treatment with lithium complex during the entire period of pregnancy led to (i) increase in the death rate of embryos and fetuses before implantation by 17.5 ± 3.1% (dose 2000 mg/kg) and after implantation by 26.6 ± 5.2% (dose 2000 mg/kg) and 15.0 ± 2.3% (dose 400 mg/kg), (ii) increase in the number of fetuses with hemorrhage on the skin by 12.4% (dose 2000 mg/kg), and (iii) pathologica changes in the internal organs on the average by 50.0% (dose 2000 mg/kg) and 30.7% (dose 400 mg/kg). In all cases, the significance level was p < 0.05. In the postnatal period of development of the offspring of animals treated with lithium complex from 6th to 20th day of pregnancy at a dose of 2000 mg/kg, the study revealed (i) decrease in the survival rate of pups by 57.7%, (ii) slowdown in their physical development rate by 32.4% and in the formation of sensory-motor reflexes by 21.7 %, and (iii) decrease in the ability to learn and adaptive behavior by 46.9% ( p < 0.05) Thus, the lithium complex exhibits dose-dependent embryotoxicity of moderate severity, manifested in both antenatal and postnatal periods of offspring development.
