Whether insulin-producing cells can be derived from non-endocrine cells and may we pharmacologically modulate conditions that will be facilitate this process is a question of great importance. It is known, that transcription factor Pdxl, product of pdxl gene, plays a key role in the development of pancreas and activation of Pdxl production may increases differentiation of non-endocrine cells into insulin + cells. Pdxl amount is affected by inflammation level and, possible, blood glucose level. Immunohistochemical study of pancreas was shown, that injections of sodium aminophthalhydrazide (APH), which modulates activity of macrophages and possess an anti-inflammatory and hypoglycemic effects, to rats with experimental type 2 diabetes (T2D) leads to the growth of Pdxl production in exocrine part of the pancreas and increase in number of extra-islet insulin + cells, both acinar and ductal. Optical density of cytoplasm in extra-islet insulin + cells rise up in APH-treated diabetic rats. An increase in the number of insulin + cells in APH-treated animals is accompanied by a decrease in their size.
