Poly(ADP-ribosy)lation is a posttranslational modification of proteins by transferring poly(ADP-ribose) (PAR) to acceptor proteins by the action of poly(ADP-ribose) polymerase (PARP). It is initiated by transferring ADP-ribose primarily to target proteins to create ADP-ribose to proteins; this reaction is followed by chain elongation and branching, resulting in the formation of a long, branched chain of ADP-ribose. PARP family is composed of 17 members with different cellular localization and activities, participating in diverse cellular functions including mitosis, transcription and cell death.
