About 4.5 million units of fresh frozen plasma (FFP) were transfused in 2008 in the United States, and these figure had increased 11% by 2010. The profile of inflammatory mediators seems to be modified by ageing, and these, along with other mediators, could participate on undesirable effects induced by transfusions. It is unknown if they influence on cellular activation. Objective. To evaluate if FFP of healthy donors grouped by ages modifies activation of U937 cells in vitro cultures. Methods. Sterile aliquots (15 ml) of FFP were facilitated by our institution’s blood bank, and were grouped depending on the age of the donors in 19-30, 31-40 and 41-50 years old. U937 cells were cultured under the following conditions without or with individual samples of FFP: 1) negative control, cellular culture in RPMI; 2) positive control, RPMI plus 1µg/mL of lipopolysaccharide (LPS) from E. coli (serotype 055:B5), 3) RPMI with 20% of FFP, 4) RPMI with 40% of FFP, 5) RPMI with 20% of FFP plus LPS and 6) RPMI with 40% of FFP plus LPS. Cytokine profile (TNF, IL-1, IL-6, IL-10) was evaluated in supernatants of cultures and in FFPs by ELISA, while the expression of activation molecules (CD11b, caspase 3 and TLR2) were measured by real time PCR; finally phagocytosis as a functional activation surrogate was evaluated by flow cytometry. Results. Production of TNF, IL-1 (p<0.01), and IL-6 by U937 only increased vs controls when 20% FFP of the 41-50 group was added to cultures. When LPS was added to cultures, an increase in all cytokines was documented (p<0.05); after a second stimulus with 20% FFP, cytokines didn’t increase further, but decreased levels of IL-6 and IL-10 (p<0.05) were seen; interestingly the only exception for these results were found with 20% FFP from the 41-50 group, which induced greater levels of TNF and IL-1 (p<0.01). Addition of 40% FFP only induced a discrete reduction of cytokines. Caspase 3 production was increased (p<0.001) in the presence of 20% FFP from 19-30 and 31-40 groups, while 20% FFP from 41-50 group increased the expression of CD11b (p<0.05). When 40% FFP was used a reduction in the expression of activation molecules was seen, except for caspase 3 in the 19-30 group. Phagocytosis was decreased in cultures with FFP from the 19-30 (40% FFP) and the 31-40 group (20 and 40% FFP). The 31-40 group was found to have the highest plasmatic levels of TNF and IL-10 in FFP, whilst the lowest were in the 41-50 group. Only in this latter group, IL-6 levels were twice as high as that of other groups. Conclusions. Only FFP of the 41-50 group didn’t decrease cellular activation, and although it induced an increase in the production of inflammatory cytokines, these FFP had the lowest concentrations of such cytokines.
