•A Tandem Liquid Chromatography and Tandem Mass Spectrometry (LC/LC-MS/MS) Technique to expedite the separation and quantification of isomers.•Chiral chromatography and reverse-phase chromatography are coupled with mass spectrometry to analyze isomers in biological matrix.•A new method to separate and quantify steroid isomers, 11ß-methyl-19-nortestosterone (11ß-MNT) and endogenous testosterone, in serum.
