•Inter- and intra-fraction seminal vesicle motion is substantial, ranging from 1 to 7 mm.•Seminal vesicle and prostate motion are weakly correlated (range R2: 0.026 to 0.7).•Current IGRT techniques require an 8 mm PTV-margin around the seminal vesicles.
