•The epigenetic drug, SAHA, sensitizes the chemoresistant colorectal (CRC) cells to 5-FU.•Sequential SAHA+5-FU treatment synergistically decreased the viability in CRC cells.•Combined SAHA+5-FU exposure increased the level of apoptosis/necrosis in CRC cells.•SAHA in combination with 5-FU also induced the level of micronuclei synergistically.
