•Paclitaxel upregulated TRPV1 in the lumber DRG and spinal dorsal horn at day14 post paclitaxel treatment.•Paclitaxel upregulated CGRP and substance P, and induced spinal dorsal horn microglia and astrocytes activation.•Duloxetine ameliorated PTX induced TRPV1, CGRP, SP upregulation, and mitigated PTX induced spinal cord glia activation.
