•Methodology for the isolation of NDMA from drug substances by SPE.•Elution profile of NDMA is affected by the properties and amount of coexisting substances.•Less than 20 mg of specimen in sample preparation for LC–MS/MS analysis.•Contrary to previous reports, NDMA was found to be stable over a wide pH range.
