人鼻病毒1B感染致人肺支气管上皮细胞代谢组改变的研究

Abstract
目的:探讨人鼻病毒1B (human rhinovirus 1B, HRV1B)感染对人肺支气管上皮细胞BEAS-2B代谢组改变的影响。方法:HRV1B感染BEAS-2B细胞6 h和12 h后,采用非靶向代谢组学技术研究BEAS-2B细胞代谢组的改变情况。结果:HRV1B感染BEAS-2B细胞6 h和12 h分别有93个差异显著代谢产物(differentially significant metabolites, DSMs)(上调47个DSMs、下调46个DSMs)和88个DSMs(上调37个DSMs、下调51个DSMs);HRV1B感染动力学(HRV1B 6 h∶12 h)过程中共鉴定到30个DSMs(上调12个DSMs、下调18个DSMs)。3组中未知DSMs占据比例均较大。脂肪酸、脂质、氨基酸、核苷酸和糖类的比例与HRV1B的感染时间呈现正相关,邻苯二甲酸二异壬酯等为HRV1B感染BEAS-2B细胞后的共同DSMs。结论:HRV1B感染致BEAS-2B细胞的脂肪酸、脂质、氨基酸、核苷酸和糖类代谢物发生明显改变。
