Abstract
•Ti/Ag NFs/Ag NPs multilayer SERS substrate was prepared via facile in-situ chemical deposition technique.•Morphology regulating of SERS substrates guiding by FDTD method, consequently fulfill strong electromagnetic field enhancement of each ‘hot spot’.•Realize high spectral resolution and super sensitive detection of small molecules and the spectrum band of PA was verified and assigned by DFT.•By using this substrate, we successfully realized the detection of UA in human urine.•Compared the surface Raman enhancement effect of different SERS substrates such as gold, silver and copper. (achieved by in-situ chemical deposition)
