Abstract
Objective: To analyze the expression of Gasdermin-D in colorectal cancer and its correlation with clinicopathological characteristics and prognosis.Methods: Seventy-eight cases of colorectal cancer tissues and paracancerous normal tissues were collected at our hospital from September 2017 to July 2019. The expression of Gasdermin-D in colorectal cancer tissues and paracancerous normal tissues was determined by immunohistochemistry. The expression of Gasdermin-D in colorectal cancer tissues and paracancerous normal tissues was determined via Western blotting. The correlation between the expression level of Gasdermin-D in colorectal cancer tissue was compared to the patients clinical data. The correlation between the expression level of Gasdermin-D and patient prognosis was analyzed using the Kaplan-Meier survival curve.Results: Gasdermin-D was mainly expressed in the cell membrane and cytoplasm, and the positive staining was brownish-yellow in color. Moreover, the expression level of Gasdermin-D in colorectal cancer tissue was significantly lower than it was in paracancerous normal tissues. There was no significant correlation between the expression level of Gasdermin-D in colorectal cancer tissues and factors such as age, gender, tumor site, tumor pathology classification, and so on. (P>0.05); however, it was clearly related to the Dukes stage, the degree of differentiation, lymph node metastasis, and the depth of the tumor invasion (P<0.05). The Kaplan-Meier survival curve analysis revealed that the median survival of the high-expression Gasdermin-D group was 51.79 +/- 4.03 months, and the median survival of the low-expression Gasdermin-D group was 36.97 +/- 9.94 months; thus, and the survival of the low-expression Gasdermin-D group was significantly lower than was that of the high-expression Gasdermin-D group.Conclusion: The expression level of Gasdermin-D in colorectal cancer tissue is significantly lower than it is in paracancerous normal tissues; its expression level is related significantly to and patients prognoses and clinical case factors such as the Dukes stage, degree of differentiation, lymph node metastasis, and depth of tumor invasion. Thus, it may be used as an important indicator to evaluate the occurrence, development, and prognosis of colorectal cancer.
