•A star polyol was developed from ε-caprolactone via ring opening polymerization.•The glucose-based polyol was used in water-based polyurethane dispersions synthesis.•Resulting three-dimensional polyurethane network has unique properties.•The star-hyperbranched polyurethane is biodegradable and biocompatible.•Amphiphilic Polyurethane is a biomaterial candidate for biomedical application.
