Anti-t2dmeffectof xiao-xian-xiongdecoction through inhibiting inflammatory reaction and reducing oxidative stress level in rats

Abstract
Objective: To investigate the effect of Xiao-Xian-Xiong Decoction (XXXD) on the inflammatory reaction and oxidative stress in type 2 diabetes mellitus (T2DM) rats. Methods: A T2DM mouse model was established by giving rats a high-glucose and high-fat diet combined with the intraperitoneal injection of STZ. The rats were randomly divided into a model group, metformin group (100 mg/kg), high-dose XXXD group (1000 mg/kg) and low-dose XXXD group (500 mg/kg), 10 rats in each group, and 10 healthy rats were randomly selected as the normal group. The rats were administered with the corresponding agents once a day by gavage for 4 weeks and relevant indicators were detected. Results: The results showed that compared with those in the model group, the contents of FBG, FINS and the value of IRI in XXXD group were significantly decreased (P < 0.01), and XXXD could improve the glucose tolerance and alleviate the pathological changes of liver tissue in T2DM rats. Compared with those in the model group, the contents of TG, TC, LDL-C, TNF-alpha, IL-1 beta, IL-6 and IFN-gamma in XXXD group decreased (P < 0.05, P < 0.01), while the contents of HDL-C increased (P < 0.05, P < 0.01). In addition, compared with those in the model group, the activities of SOD, GSH-Px and CAT and the expression of p-FoxO1 protein were increased in XXXD group (P < 0.05, P < 0.01), while the MDA content and p-Akt protein expression were decreased (P < 0.05, P < 0.01). Conclusion: These results indicate that XXXD has an anti-T2DM effect in rats, which is related to its inhibiting inflammatory reaction and reducing oxidative stress level by regulating PI3K/Akt/FoxO1 signaling pathway.
