The few data regarding ischemic preconditioning (IPC) of the small bowel encouraged us to investigate its effect on signal transduction mechanisms in bowel mucosa cells. In 10 mongrel dogs the intestine was preconditioned by 4 cycles of 5-min ischemia, 10-min reperfusion of the mesenteric artery was employed. Activity of transcription factor, nuclear factor-kappaB (NF-kappaB) was detected in mucosa cells by gel electrophoretic mobility shift assay (EMSA). Our results showed the presence of NF-kappaB in control samples, but we observed gradual activation of NF-kappaB peaking by I hour, and falling 3 hours past preconditioning. Our findings confirmed a time-dependent activation of NF-kappaB in bowel mucosa after IPC.
