Network Pharmacological Study Of Effectiveness Of Cannabis Sativa L. Against Orthopedic Trauma

Orthopedic trauma is a medical condition depicting most common bone injuries which affect individuals of every age and gender worldwide. These conditions can pertain to bone fractures, musculoskeletal injuries, and blunt trauma. Cannabis sativa L. is a primitively used plant in both medicinal and industrial uses. However, its mechanism of action is not fully investigated in the case of orthopedic trauma. The aim of the study was to determine and elucidate the potential of C. sativa in managing orthopedic trauma. The study spanned over a comprehensive literature survey which aimed to determine the interaction of bioactive compounds of C. sativa, followed by the screening of various target compounds relative to orthopedic trauma. The Gene Ontology (GO) term enrichment analysis was also conducted using Cytoscape software and its plugin, before which the PPIN was constructed with the help of STITCH, respectively. The results of the literature survey demonstrated 13 components in C. sativa, which yielded the presence of 101 potential targets and 12 relative targets to C. sativa. These potential targets were then screened for their association with the management of orthopedic trauma. The protein-protein interaction network revealed the association of several protein targets such as IL6, FOS, JUN, and PTGS2. Moreover, 4 GO terms were also elucidated using enrichment analysis which were evaluated using gene functional annotation clusters and their abundance values, respectively. The results of the study revealed the potential mechanistic action of C. sativa in the management of orthopedic trauma, which is demonstrated via the in silico analysis that it occurs through the help of interacting with various biological pathways via target proteins. The observed mechanisms and biological processes can be the regulation of extrinsic apoptotic signaling pathway via death domain receptors, fatty acid derivative biosynthetic process, eicosanoid biosynthetic process, and unsaturated fatty acid biosynthetic process, respectively.
